Abstract

This project is designed to provide all research groups within the SFB with standardized protocols, technical assistance, ultrastructural analysis and material from human skin disease. This includes standard procedures for primary cells and obtained from mouse or human skin, and the generation of complex in vitro systems involving different cell types. The success of experiments using primary cells often depends critically on small variations of the experimental conditions. To achieve high comparability of results obtained by different groups we need standardized procedures, which will be provided by this project. Histopathological expertise will be provided to the different groups for both mouse and human biopsies. This includes the maintenance and expansion of a tissue bank obtained from patients and from mouse models (e.g. tissue repair following wounding). In our previous work we have already established such a database/tissue bank and this will now be expanded to include the tissue banks collected in the three clinical networks coordinated by the Department of Dermatology. This bank and its expansion will be instrumental in translating our findings to the clinic.

Kurzbeschreibung

Ein wichtiger Teil dieses SFBs ist die Analyse von genetisch veränderten Mausmodellen. Die genaue phänotypische Charakterisierung der Haut dieser Mäuse erfordert eine hohe Expertise in unterschiedlichen Techniken der Zellbiologie, Histologie und Elektronenmikroskopie. Wichtige Aufgabe vieler Projekte innerhalb dieses SFBs ist darüber hinaus die Übertragung der an Mausmodellen gewonnenen Erkenntnisse auf die humane Situation. Daher muss eine entsprechende Gewebebank mit Proben von Erkrankungen der Haut analysiert werden. Dieses Z2 Projekt soll die hierzu notwendige Expertise für die Isolierung von primären Zellen, die histologische Analyse von Mäusen und humaner Haut, sowie deren pathologischen Veränderungen unter standardisierten Bedingungen gewährleisten.
